Process: The semen is layered over density gradient media in a centrifuge tube. It is spun at high speed, separating sperm by weight. The most motile, morphologically normal sperm settle at the bottom and are collected. ✔ Pros: Highly effective for selecting high-quality sperm. ❌ Cons: Requires centrifugation, may cause slight sperm DNA damage. 3️⃣ Simple Wash Method (For IUI & IVF) Best for: Cases where only mild processing is needed. Process: Semen is mixed with a washing medium and centrifuged. The sperm pellet is re-suspended in a fresh medium. ✔ Pros: Quick and simple. ❌ Cons: Less effective at selecting highly motile sperm. 4️⃣ Magnetic-Activated Cell Sorting (MACS) (For IVF & ICSI) Best for: Removing apoptotic (dying) sperm with DNA damage. Process: Uses magnetic beads that bind to apoptotic sperm, removing them from the sample.
